Although the technology of discovering molecular markers based on Next Generation Sequencing (NGS) is very mature, it is still time-consuming, expensive and challenging to conduct high-throughput data analysis, especially for plants with high heterozygous genome characteristics. Compared to NGS, the advantages of DNA microarray are fast, accurate, and inexpensive. And more advantages are: (1) DNA microarray for polyploid genotyping is far better than NGS; (2) The mutation sites of DNA microarray are fixed. In the following GWAS research in breeding and transformation applications, DNA microarray is easier to standardize; (3) Compared with NGS, DNA microarray results are stable, fast and easy to analyze. The results can be used for quantitative trait loci (QTL) analysis to identify how the gene expression leads to phenotypic differences.

DNA Sample requirements

  • Amount: no less than 1 ug for each sample;
  • Concentration: 0.5-50ng/µl;
  • Purity: OD 260/280 should be 1.7 ~ 1.9;
  • Quality: genome should be integral, no degradation, no RNA contamination or other genomic DNA contamination;
  • Preservation and transportation: samples should be transported at low temperature (- 20℃) and sealed with sealing film to prevent contamination.
