MicroRNA (miRNA) is a kind of endogenous non-coding RNA with regulatory function found in eukaryotic organisms. It does not have an open reading framework (ORF) nor encode proteins. The miRNA is composed of 20-24 nucleotides. The precursor has a predictable stem ring structure. The miRNA is highly conservative with time-sequential and tissue-specific expression. It guides the RNA-induced silencing complex (RISC) to degrade or inhibit mRNA translation by pairing with target genes, thus regulating gene expression at the transcriptional or post-transcriptional level1. MicroRNA is involved in many physiological processes, such as cell proliferation, differentiation, apoptosis, tissue and organ formation and disease occurrence. Abnormal expression of miRNA often leads to disease.
CD Genomics' miRNA profiling microarray can specifically detect mature microRNA and distinguish highly homologous microRNA molecules, which can be further applied in scientific research and genetic engineering. Our miRNA microarrays are mainly used to detect the expression level of microRNAs, screen differentially expressed microRNAs and predict differentially targeted microRNAs. The probe information of our miRNA profiling microarray products comes from the Sanger MicroBase database, which contains microRNA detection probes for human, rat, mouse, animal, and plant species, covering mature microRNAs and pre-microRNAs. They have the characteristics of complete species, high coverage and reliable results.
Because there are more than 20 bases of miRNAs, which are relatively conservative, highly homologous, precursors coexist with mature microRNAs and wide dynamic expression range, the detection of microRNAs is facing great challenges. The miRNA profiling microarray of CD Genomics has a special probe design. Each probe is repeated at least 20 times to ensure that only mature microRNAs are recognized. The microarray is highly sensitive and can identify each base difference with a detection limit of less than 0.1 amol. The minimum sample size for detection is only 100ng total RNA. There is no need to separate the miRNAs to avoid errors. The dynamic detection range is wide and the detection abundance spans 5 logs, which is conducive to the detection of a wider range of miRNAs. The average CV was less than 10% and R² was more than 0.99. It can also be detected in plasma, serum, body fluids and exosomes with low expression abundance.
